The goal of a study was identification and comparison of the secretion of EGF-like growth factors, the level of nitric oxide production and expression of the estrogen receptors under the influence of T-lymphocytes and in the control samples in different cell models. To achieve the goal used cytological and immunocytochemical staining, method of Griess, electrophoresis in polyacrylamide gel, Western Blott analysis. In the first phase of the study was extended a study of correlation between the level of intracellular nitric oxide, proliferative activity of cells and the effect of T-lymphocytes in these processes in 2-D and 3-D cell cultures. It was found that the level of intracellular nitric oxide positively correlated with the degree of cancer progress. The presence activated donor T cells and MT-4 cell's cytokines and activation of the iNOS changes the intensity of NO generation by epithelial cells in cell medium during cocultivation. It leads to the inhibition of apoptosis in all samples. Increase of NO levels, decrease of apoptosis and activation of ER at the same time indicates poor prognosis of cancer progress. In the second stage of the study was shown and compared production of EGF-like peptides in monolayer and spheroid cell models. It was calculated the number of EGF-like proteins on 1 million cells by peak areas, staining intensity and cells number in cell culture. It was found that cells of monolayer secrete into the culture medium is 2.5 times more EGF-like proteins than cells of spheroids.
