Performed clonal analysis for five producers from "Collections" SI IFBG NAS of Ukraine, which produced 6-12 g/dm3 of lysine. It was established that the investigated clones remained auxotrophic for leucine and become auxotrophic for threonine, methionine, tryptophan, and homoserine. Performed mutagenesis using NTG and UV, new clones producing strains was obtain. The new M-strains were selected from regulatory and analog rezistent auxotrophic as genetic markers. Vector systems and the schemes of transformation were chosen for gene pyc, lys C, dapA. The optimum conditions for the cultivation of new M-strains were selected. The biosynthetic activity of these M-strains for lysine (50-60 g/dm3) and conversion rate of sugar (45%) under conditions optimized were defined on laboratory equipments.
