Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology popup.description2 The level of DNA damage in ovarian surface epithelial (OSE)cells derived from women without family history of cancer (control) was 1,0±0,2 and significantly increased to 15,1±0,1 in OSE derived from women at high risk of developing ovarian cancer. Densimetric analysis of vestern-blot of control OSE has shown that optical density was 0,04 for FANCD2-L and 0,8 - FANCD2-S, which indicates about presence of monoubicvitinated (FANCD2-L) and nonubicvitinated (FANCD2-S) FANCD2 protein. In OSE of women at high risk of developing ovarian cancer optical density FANCD2-L significantly increased to 0,002 and FANCD2-S - to 0,2. р53 expression was significantly higher in OSE of women at high risk of developing ovarian cancer (optical density 0,9) compared to control cells (0,001). These results indicate OSE of women at high risk of developing ovarian cancer has significantly higher DNA damage compared to control cells, probably due to lost of key reparation protein FANCD2-L.
