We made theoretical models of specific interaction of cytochrome P450 2E1 with ligands, which were allowed to explore the topological characteristics of the active center of the enzyme. Conditions for heterologous expression, isolation and purification of recombinant functionally active cytochrome P450 2E1 were optimized. The amino acid residues important for selective binding of the active site of cytochrome P450 2E1 with substrates (Phe 298 and Thr 212) and inhibitors (Phe 106 and Phe 298) were found by using theoretical models. The computer models data of criticality of amino acid residues Phe 106, Phe 298 and Thr 212 for the functioning of cytochrome P450 2E1 were experimentally verified. These results have a great fundamental and practical importance for enzymology, pharmacology, diagnostics and medicine.
