The development is based on the study of the physical parameters of complex formation in a multimodal and competitive binding of biologically active substances. The research was carried out by spectrophotometry, fluorescence and vibrational spectroscopy, differential scanning calorimetry (DSC), EHF dielectrometry, and computer simulation using the polynucleotides, DNA and biologically active substances (antitumor antibiotics and their newly synthesized analogues, mutagenes, and vitamins). New information on the types and structures of DNA - ligand complexes and their thermodynamic characteristics in multimodal and competitive binding taking into account the water environment was obtained. A new technique for calculating the parameters of ligand binding to the DNA according to the DSC was developed. The method for determining the effect of various substances on the structure of DNA was developed and patented. It allows to perform a preliminary assessment of the level of BAS mutagenicity. Online database ProtNA-ASA (www.protna.bio-page.org) was created. It provides systematic information about the conformational parameters and areas of accessible surface of atoms of nucleic acids in the protein-nucleic acid complexes.
