Research methods - morphological, histological, spectrofluorimetrical and techniques of monolayer and three-dimensional in vitro cultivation. The studies were carried out on cultures of cryopreserved mesenchymal stem cells (MSC), isolated from human adult bone marrow (BM) and adipose tissue (AT). Novelty of the results - Using qualitative and quantitative methods the complex research of the efficacy of adipogenic and osteogenic differentiation of bone marrow and adipose tissue mesenchymal stem cells was carried out. As a result, with the application of quantitative methods for the determination of MSC differentiation products the significant differences in the efficiency of induced adipo- or osteogenesis between cell cultures, derived from bone marrow and adipose tissue was not revealed. It was first shown that the three-dimensional cultivation within alginate-gelatin scaffolds promote both the adequate realization of the proliferative potential of MSC cultures and maintenance of the ability of these cells for induced differentiation into adipogenic and osteogenic lineages. The results obtained allows making a significant contribution to the knowledge of the potential and properties of mesenchymal stem cells, derived from different adult human tissues during monolayer culture in vitro as well as within three-dimensional polymer carriers.
