The purpose of the first phase of work was to identify and fix the control kinetic parameters of growth spheroid' and monolayer' cell cultures of breast cancer cells. As these indicators were selected following parameters: the intensity of proliferation of tumor cells, the ratio of alive and dead cells in culture, the cells sensitivity under the influence of external factors, the number of cells which were attached to culture surface, distribution of cell populations for cell cycle phases, the level of expression of epidermal factor growth and pro-apoptotic protein p53. Methods: cell culture, cytological, colorimetric, biochemical and statistics. Results: The majority of indicators have been identified on the models of spheroid' and monolayer' growth, used methods of cytological and immunocytologycal staining, colorimetric and cyto-fluorometric study of cell populations, electrophoresis. It was found that monolayer sensitivity to external factors significantly higher than spheroid, and the survival rate is lower. Proliferative potential of cells depends on cultivation conditions, such as characteristics of the substrate. Adhesion to the substrate and cell proliferation have direct contact and describe the intensity of the tumor process. At the stage of exponential growth measured by cell division phases of cell cycle by cytofluorometric and cell migration assaies were found between the suspension and adhesive fractions of monolayer and spheroid cultures. The expression of p53 and EGFR were measured in monolayer culture of MCF-7 cells.
