The object of research: tumor cells of breast cancer MCF-7 of monolayer and spheroid growth and T-lymphocytes (line MT-4 cells of T-cell chronic lymphocytic leukemia human donor T lymphocytes). Objectives of the study was to determine and compare the cellular models of epidermal growth factor-like growth factors secretion , the level of nitric oxide production by cells, and expression of estrogen receptor in both control and under the influence of T-lymphocytes. To solve the problems was used cytological and immunocytochemical coloring technique, Grissa-assay, electrophoresis in polyacrylamide gels, Western BlotT. The first phase extended the study of correlation of the level of intercellular nitric oxide, cell proliferation and influence of T lymphocytes in these processes in 2-D and 3-D cultures. It was found that the level of intercellular NO, has a direct correlation with the degree of tumor process. Tumor cells cultivation in presence of cytokines from activated donor T cells and MT-4 leaded to activation of iNO-synthase in the culture medium. It stimulated the formation of nitric oxide by epithelial cells. This in turn leaded to inhibition of cell apoptosis for all samples. As a result a greater level of NO, reduction of apoptosis and activation of estrogen receptor can conclude that adverse weather flow of the tumor process. The second stage of work was in determination and comparison of rates of production EGF-like peptides in monolayer' and spheroid' cellular models. The area of color peaks, the intensity of staining, the number of cells in culture was calculated production of EGF-like proteins at 1 million cells. It was found that the tumor cells in culture monolayer culture secrete in 2.5 times more EGF-like proteins than cells in spheroid.
