The aim. The estimation of the state of epithelium of tumor- distant oral mucosa by the complex of histostructural and cytological index, expression of p53, Ki-67, Всl-2 proteins and content of DNA in patients with oral squamous cell carcinoma (OSCC). Materials and methods. The biopsy specimens of tumor-distant oral mucosa and tumors were obtained from 18 patients with OSCC (male, age from 43 to 79 years) The biopsy specimens of oral mucosa and tumors were classified according to the UICC criteria. Tissue sections were immunohistochemically stained using monoclonal antibodies: for p53 (clone DO-7,antibody to wild/mutant type), or Ki-67 (clone МІВ-1) for Всl-2 (clone 124), and En Vision "Daco Cytomation" (Denmark) and counted according as Labeling Index (LI %). DNA content was analyzed using Feulgen staining and estimated by cytospectrophotometry. Results. Most of the patients with OSCC (83,3%) have the smoking-drinking status and long-term professional contact with carcinogen and mutagen. In tumor-distant oral mucosa revealed the progression of histopathological phenotype to hyperplasia(2), to dysplasia mild (2), dysplasia hight (3) to carcinoma in citu (4), to invasive carcinoma (7). The results showed significant variation of expression p53, Ki-67 and content DNA in cells of tumor-distant mucosa. Labeling index expression of p53, Ki-67 in tumor-distant oral mucosa and mean percentage of aneuploid cells were higher in the severe dysplasia, Ca in citu. Its can be used for differential diagnostics of dysplasia and cancer. Conclusion. Expression proteins p53 and Ki-67 in epithelial cells and aneuploidy are the indicator of the transformation phenotype in tumor-distant oral mucosa and represent high risk of development of second primary carcinomas and recurrent after treatment.
