Investigation of the expression of protein markers of the epithelial stem /progenitor cells p63, CK5/14 , of biomarker p53 in epithelial layers of tumor-distant oral mucosa in patients with oral squamous cell carcinoma (OSCC). Materials and methods. The biopsy specimens of tumor-distant oral mucosa and tumors were obtained from 18 patients with OSCC (male, age from 43 to 79 years). The alteration of oral mucosa and tumors were classified according to the UICC criteria. Immunohistochemically were used expression of regular nuclear marker of the epithelial stem /progenitor cells p63 and marker keratin CK5/14 of basal cells, biomarker protein p53 in epithelial layers of tumor- distant oral mucosa and tumors. Tissue sections were immunohistochemically stained using monoclonal antibodies: for р63 (clone 4A4, antibody to all isotype ),for CK5/14 (clone124 ) and En Vision "Daco Cytomation" (Denmark) and counted according to the epithelial layers as Labeling Index (LI %). Results. Most of the patients (83,3%)15/18, і 20 years і 20 cigarettes per day) have the smoking tobaсco -drinking status and long-term professional contact with carcinogen and mutagen. The positive expression of markers p63, CK5/14, p53 were detected in all carcinomas (OSCC),( range p63 LI(%) 19,7%- 79,1%; range LI(%) CK5/14, 8,4%-54,3%) .Its demonstrate the participate of cells that expression of marker proteins of epithelial stem/progenitor cells p63 and keratin CK5/14 in biology of OSSC. The analysis showed the architectural alteration of cells p63 and CK5/14 expression in the epithelial layers of tumor-distant oral mucosa and distribution in suprabasal layers. The suprabasal expression of p63, and CK5/14 correlated with grade dysplasia and carcinoma in citu in tumor-distant oral mucosa. Labeling Index (LI %) of p63 and CK5/14 decrease in basal layer and increase in suprabasal layers from mild dysplasia to height dysplasia ( dysplasia height range LI(%), basal layer, p63 12,0%-30%, СК5/14,16,0%-23,0%;suprabasal layers, p63, 34,6%-70,0%, СК5/14, 25,0%-34,0,%) carcinoma in citu and invasive carcinomas in tumor-distant oral mucosa. Conclusion. That results indicated that the marker stem/progenitor cells of basal layers proteins p63 and CK5/14 including participate in formation of transformation phenotype. The immunohistochemicall Labeling index(LI%) of p63, CK5/14, p53 cells in suprabasal epithelial layers of oral mucosa can be used for differential diagnostics of grade of epithelial dysplasia and carcinoma in situ and monitoring of oral mucosa after treatment.
