The object of study - plasmid DNA, recombinant proteins, plants, transient expression. The main goal of the research - was to create an effective genetic constructs for production of proteins ESAT6 and Ag85В playing a significant role in the immune response in tuberculosis, and human interferon alpha 2b and obtaining the transgenic model, edible and fodder plants with high levels of expression of genes encoding highly immunogenic proteins of causative agent of tuberculosis and antivirus proteins. At this stage of the goal was to create and analyze the genetic constructions of genes encoding proteins stimulators of immune response in tuberculosis, and the gene for leukocyte interferon highly effective expression in plants, as well as the development of techniques for plant transformation vectors containing the genes encoding ESAT-6, Ag85B and interferon. Methods of research - cloning of DNA sequences, polymerase chain reaction, genetic transformation, transient expression of recombinant proteins in plants, separation of DNA and proteins using electrophoresis, Western immunoblotting and chromatographic purification of proteins. As a result of the research two sets of vectors was created - for Agrobacterium-mediated transformation of plants and for expression of target proteins in a prokaryotic system. For the nuclear transformation of plants binary constructions was created pCB064, pCB067 and pCB158, containing in the same reading frame sequences of esxA and fbpBdTMD genes, encoding antigens of Mucobacterium tuberculosis ESAT6::Ag85B (dTMD), and vectors pCB124, pCB125, pCB161 and pCB196, containing the gene for interferon human alpha 2b. To construct vectors for expression of target proteins in a prokaryotic system sequence fused genes esxA:: fbpBdTMD, encoding the antigens ESAT6::Ag85B (dTMD), and interferon gene human alpha 2b were cloned into appropriate expression vectors.
