It also takes part into the different physiological and pathophysiological processes in human organism. Plasmin, the key enzyme of fibrinolytyc system, possesses different regulator functions. It can make influence on the activity of other enzymes, growth factors and cellular receptors. So, investigation of plasminogen/plasmin activity in haemostasis system under the normal and pathological state is an issue problem. The main tasks of the work concern the study of activity regulation of plasmin by the fragments of fibrinogen and fibrin, the investigation of relationship between platelets and plasminogen/plasmin system and the development of quantitative methods for determination of the main components of fibrinolytic system. We have established that early products of fibrin degradation inhibit the rate of clot lysis; DDE, DD, Е1Е2 and Е-early fragment of fibrinogen stimulate the reaction of plasminogen activation by tissue plasminogen activator, but they can't defend plasmin, which is formed on their surface, from the inhibition by ?2-antiplasmin. So, accumulation of fibrin degradation products during clot lysis can be the cause of clot resistance towards hydrolysis. It was shown that platelets can secrete an active form of PAI-1 during aggregation. The forming PAI-1 inhibits tissue plasminogen activator. Resting platelets facilitate plasminogen activation by tissue plasminogen activator. Lys-plasminogen inhibits platelet aggregation induced by thrombin and collagen. We have developed the methods for determination of plasminogen concentration, tissue plasminogen activator and PAI-1activity with the usage of fibrin as both stimulator and substrate for reaction of activation and proteolysis. The approbation of these methods has been carried out at different pathologies. The methods can be used to estimate blood fibrinolytic potential in clinicodiagnostic laboratories. The results of the carried investigation expand our understanding about the regulation of haemostasis system as a whole and existing relationship between its separated parts.
