In our experiments, the amplitude of calcium transients induced by KCl depolarization of the membrane in neurons after incubation of cells with beta1-42 amyloid increased by 37% compared with the control. Application of memantine led to a decrease in calcium loading endoplasmic stores and reduce the amplitude of the calcium transient depolarization of the membrane KCl, but did not change the level of free calcium. This effect was more pronounced in cells incubated with 1- 42 beta-amyloid. Dantrolene, ryanodine receptor antagonist, had a similar effect, but it reduced free calcium by 10% in cells with beta1-42 amyloid in comparison with the control.
