The aim of investigation was to find the connection between the children brain tumor cells individual sensitivity to the drugs and the expression levels of apoptotic genes, genes, that are responssve for the tumor cells adaptation to the hypoxia and genes, that determine the differentiation grade. To estimate the drug efficiency by investigation of brain tumor nestine-positive reduction under it's influence during short-term culturing. Methods of the investigation: short-term culture, polymerase chain reaction with reverse transcription, immunocytochemistry. In the brain tumor operative materials of children until 16 years old the expression of genes СD133, HIF-1alpha, MDM2, BAX, BCL-2, SOX2, Nanog, Oct3/4 was determined in the suspension of native tumor cells and after short-term culturing with the drugs. It was determined, that relation between expression level of BAX : BCL-2, expression levels of HIF-1alpha, СD133, BCL-2 and MDM2 in the native cells and after the culturing are the informative prognostic factors of the children's brain tumor drug sensitivity to the lomustine (CCNU) and temozolomide (TMZ). The method of short-term culturing in the presence of drugs, after caring out clinical trials, may be recommended for the anticancer therapy protocol choice.
