Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology popup.description2 Ovarian surface epithelial (OSE) cells from women at high risk (HR) of ovarian cancer (OC) are characterized by alteration of structural integrity of DNA and the level of relative tail moment of DNA-comets was in 15 times higher compared to control cells. After immortalization of OSE cells with E6/E7 HPV 16 oncogenes the 3-fold increasing of DNA damage level has been revealed in control cells as well as in cells from women at HR of OC. Cell treatment with mitomycin C (MMC) caused progressive increase of genome alterations of OSE cells from women at HR of OC, whereas control cells were resistant to MMC. Immortalized by HPV oncogenes control and HR OSE cells were significantly sensitive to MMC. The high level of spontaneous DNA damage of OSE cells from women at HR of OC and low expression of ubiquitinated FANCD2 protein increase sensitivity of ovarian cells to genotoxic factors and can be background for genetic instability. HPV oncogenes in OSE cells increase the level of DNA damage, genotoxic effect of MMC and can be additional risk factor of malignant transformation of OSE.
