The aim of study: to develop scientific approaches to obtaining of safer diphtheria and tetanus toxoids without unprotective fractions and toxic modifiers. Methods of investigation: microbiological, immunological, biological, chromatographic, statistical. It was found experimentally that purified industrial tetanus toxoid contained ballast proteins, which might be shed by certain modes of ultrasonic treatment. The possibility of obtaining of diphtheria toxoid using chemical and physicochemical factors was studied. It was proved that the influence of the physical factor did not cause destruction or inactivation of native diphtheria toxin (DT). It was determined that twofold reduction of formalin percentage did not significantly affect the specific activity of obtained DT derivatives and DT derivative obtained by physicochemical factors was safe. It was established that the toxoiding process could be controlled by fluorometry. The catabolic activity of microorganisms was studied by the amount of oxidized glucose. Determination of catabolic activity of microorganisms makes it possible to conduct selective search of strains and evaluate epidemic situation.
