Significant inhibition effect of complexes at micromolar concentrations (up to 20 uM) was observed. Some compounds at concentration 5 uM and below inhibited another DNA-synthesizing enzyme, Taq-DNA-polymerase. The most active inhibitors of both enzymes were found to be monomeric and bis-clathrochelate complexes containing hydrophobic fragments of mercaptobenzoic acid. It was shown that the removal of inhibitors from the reaction mixture after the first step of analysis (i.e. telomerase reaction) is required to get correct results of the determination of inhibitors activity in TRAP system. This is due to the fact that some compounds are able to inhibit Taq-DNA-polymerase at the second step of analysis (PCR reaction) while not being telomerase inhibitors.
