Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology popup.description2 At the 3rd stage of our research, we have studied expression patterns of mRNA of mouse beta-defensins (mBD-1-4) in B16 melanoma cells in vivo. Using semiquantative RT-PCR analysis it has been shown that B16 melanoma cells express mRNA for mBD-1and mBD-2, but do not express mBD-3 and mBD-4. Administration of vitamin D3 to tumor-bearing animals caused some retardatiom in primary tumor development and had insignificant effect on B16 metastasis, and did not affect expression levels of mBDs. In 2012 we have performed cloning of human beta-defensin-4 gene (hBD-4) in bacterial cells using GST-system for production of recombinant proteins. The sudy of biologic activity of rec-hBD-4 has shown that this peptide affect viability and migration capacity of cultured A431 cells in a concentration-dependent manner, possesses antimicrobial activity and has no effect on pigmentation of cultured human melanoma cells.
