Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology popup.description2 The work is devoted to investigation of mechanisms which provide a survival of tumor cells under metabolic stress, and also to an experimental justification of ability of oxyresveratrol to inhibit growth of malignant tumours by updating of the response of tumor cells upon metabolic stress. It is established, that tolerance of Lewis lung carcinoma cells to antimetabolic/antiangiogenic therapies can be caused by a high ability of these cells under nutrient deprivation to pass in a resting phase. At the initial stages of tumor cell growth under deficiency of nutrients inhibition of autophagy with its further intensification under lactate accumulation is observed. Lactic acidosis promotes a survival of tumor cells under deficiency of glucose due to decreasing in speed of glucose consumption by tumor cells, and also due to suppression of functional antitumor activity of tumor-associated macrophages. It is shown, that high antimetastatic activity of oxyresveratrol is caused substantially by increase of antitumor activity both tumor-associated macrophages, and blood phagocytes and correlates with its significant antioxidant activity concerning tumor cells, including the conditions of deficiency of nutrients. The obtained data are theoretical base for working out of new principles of antitumor antimetabolic therapy.
