Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology popup.description2 The patterns of hBD-2 expression in human lung adenocarcinoma A549 cells and epidermoid carcinoma A431 cells have been analyzed. It has been shown that induction of expression of native hBD-2 in А-549 and А431 cells upon the action of vitamin D3 metabolites 1,25-(0H)2D3 and 25-(0H)D3 results in suppressed cell proliferation. It has been demonstrated that recombinant hBD-2 at the concentrations of 10 to 1000 nM causes suppression of proliferation of A549 cells via blockage of cell cycle in G1/S checkpoint, activation of retinoblastoma protein pRB and down-reguation of cyclin D1 expression along with nuclear translocation of the defensin. With the use of pull down assay on affine carrier hBD-2-sepharose and following mass-spectrometry analysis there have been identified potential partners of the defensin, in particular, nuclear proteins Dim1-like protein, heterogenous nuclear ribonuclear protein, methyl-CpG-binding-domen-protein-2, acidic nuclear phosphoprotein pp32, retinoblastoma-like protein 1, and membrane protein RAGE receptor. The results of experiments on formaldehyde cross-linking points on the possibility of direct interactions between the defensin and chromatin in cell nuclei. Expression patterns of hBD-2 have been studied in the samples of resected human lung tumors. There has been revealed a dependence between expression of hBD-2 peptide and differentiation grade of lung adenocarcinoma - low differentiated tumors differ from moderately differentiated ones by significantly lower hBD-2 expression.
