Aim - determination the functional and metabolic disturbances in the heart of ovariectomized female rats with insulin resistance syndrome of different genesis and searching of biologically active compounds for them correction. Methods - pathophysiological, biochemical, spectrophotometric, pharmacological, electrophysiological, immunological, organic synthesis, physic-chemical analysis. It was established that metabolic syndrome induced high-fructose diet in female rats with estrogen deficiency leads to disturbance functional state of the cardiovascular system through the development of mitochondrial dysfunction and oxidative stress in cardiac myocytes. Prolonged use of a compound PEО607, like a 17beta-estradiol prevents the development of oxidative stress in the mitochondria of cardiomyocytes, improves bioenergetic processes in the heart muscle, helps restore production oxide nitrogen, inhibits the expression of immune dysfunction and hemocoagulation in ovariectomized rats with metabolic syndrome induced high-fructose diet. Short use of a compound PEО607 reduces low-intensity chronic inflammation, improves blood rheology, slow autoimmune processes, positive effect on the functional state of the cardiovascular system, and, in contrast to 17beta-estradiol improves glucose intolerance, and insulin sensitivity in ovariectomized rats with metabolic syndrome induced by high caloric diet with thyreostat. The revealed selective effect of compound PE0607 on major factors of cardiometabolic risk justify the its perspectives as potential pharmacological agent for the treatment of metabolic syndrome in postmenopausal women with hypothyroidism.
