It is shown that carbohydrate chain length and the presence of the hydroxyl group in 11 position of the aglycone are the key factors that determine the antitumor activity of landomycins. In particular, the loss of one carbohydrate residue reduces the anti-neoplastic activity of landomycins and their ability to produce reactive oxygen species by 60 % and the elimination of the C11 -OH group leads to further reduction of these values by 30 %. This directly affects the ability of these antibiotics to produce hydrogen peroxide in the mitochondria of malignant cells and to kill drug-resistant tumor cells. These results indicate on significant prospects of the most active members of landomycin family, in particular landomycin A, in the treatment of drug-resistant tumors.
