Department of psychiatry popup.description2 Biological activity of 200 N- and S-substituted azaheterocycles compounds were investigated in the experiments in vitro and in the conditions of cerebral ischemia modeling. The most active 50 compounds were selected and their influence on antioxidative activity, state of thiol-disulfide system and power exchange of neurons were studied while adding them into suspension of cytotoxic compounds. The influence of 50 compounds on expression of inducible, neuronal and endothelial NO- synthase and on the indexes of mitochondrial dysfunction was revealed and their ability to prevent development of mitochondrial dysfunction, oxidative stress and violation of power exchange in conditions of experimental cerebral ischemia was revealed. Capacity of 50 N- and S-substituted azaheterocycles compounds to model expression of endogenous factors of cytoprotection (HIF and HSP-proteins) and to influence on the behavioral reactions of rats with modeled cerebral ischemia was defined. Toxicological parameters, LD50, LD100 were determined for 50 N- and S- substituted azaheterocycles compounds. Influence of 50 N- and S-substituted azaheterocycles compounds on morphofunctional state of neurons (density and area of neurons, their RNK content, density of apoptotic and destructively changed neurons, index of amount of the lost neurons), on early reacting genes expression in the neurons (c-fos) and content of antyapoptotic bcl-2-protein were studied. Comparative description was conducted among N- and S- substituted azaheterocycles compounds and the most active one was selected. After investigations declarative documentation on creation of new neuroprotective and antiischemic drug was worked out and submitted to State Expert Center of Ministry of Health of Ukraine.
