Aim of the work was to compare the functional activity of the іdo gene in fetal liver mesenchymal stem cells (MSCs) after cryopreservation by different regimens. Research methods were immunomagnetic cell sorting, cytofluorimetric and RT-PCR analysis. Novelty of the results. The fact of different levels of ido gene expression depending on fetal liver gestation term was established. It was noted the same trend of these changes in the total FLC suspension and separated CD105+ fraction (MSCs). The results suggest that CD105+ cells (MSCs) are the main producers of the IDO enzyme in the fetal liver. Changes of the ido expression level in total FLC suspension and in isolated fractions after cryopreservation were established. The highest increasing of ido expression was observed in the CD105+ fraction (MSCs) after cryopreservation by regimen 1. The results of these studies will allow to optimize the approaches to improve treatment of autoimmune diseases (AIDs). Results of this research can be used to decode the mechanisms of cryopreservation influence on the functional state of the genetic apparatus of cells in different types of biological objects, as well as provide an opportunity to prove the feasibility and features of cryopreserved MSCs in clinical practice, including the treatment of AIDs.
