According to the literature it is interferon alpha is one of the factors of humoral interaction of cells of mesenchymal origin with the microenvironment in the body. Object studies: interferon alpha and tumor cell lines MCF-7 and HT29. In this paper have been involved imunotsitohimicheskie, flow cytometric, molecular biological, biochemical and statistical analysis methods. The aim of the study was to characterize the effect of interferon alpha on proliferation, survival, receptor profile and the ability to form multicellular spheroids of tumor cells in vitro. The objectives of the study was to determine the kinetic parameters of the growth of human tumor cells in culture with different concentrations of interferon alpha, the evaluation level expression of some markers tumorigenic tumor cells under the influence of alpha interferon and a comparative analysis of the above parameters at different times in monolayer culture and spheroid culture. The comparison was performed on the expression level of estrogen receptors, epidermal growth factor receptor, cytokeratins and E-cadherin. The results found that MCF-7 react to the presence of high doses of cytokines IFN-alpha-2b reduced proliferative and survival both in suspension and in adhesion. When cultured with IFN-alpha-2b in concentrations of 105 IU / ml, the number of dead cells is more than twice than in the control. Direct effect of IFN-alpha-2b reduced density cell growth in culture. The antiproliferative effect of IFN-alpha-2b is already apparent after 3-4 days of cultivation. The level of expression of nuclear estrogen receptor ERalfa suppressive effect of high doses (104 and 105 IU / ml) IFN-alpha-2b. Increased the percentage of cells in culture, which do not express ERalfa. Also several levels of decreased expression of epidermal growth factor receptor. But the increased expression of differentiation markers - cytokeratins and E-cadherin. Furthermore, in the presence of interferon alpha observed dose-dependent inhibition of the generation of multicellular tumor spheroids.
