In the work we used imunocitochemisrtrycal, flow cytometric, molecular biological, biochemical and statistical analysis methods. The aim of the study was to characterised the influence of humoral factors of the microenvironment on the proliferation, survival, metastasion potential of tumor cells in multicellular spheroids. Determination proliferative and morphological characteristics of the spheroids and monolayer cultures under various conditions microenvironment. The objectives of the study was to generate a multicellular tumor spheroids (MTS) breast cancer cells. Determine the proliferative and morphological characteristics of MTS under different conditions of the microenvironment. As a result, performance was demonstrated that the cells have a greater MTS proliferative capacity and greater survival as compared to monolayer culture, when used in the experiment all the humoral effects. Thus, the number of living cells in MTS significantly higher in cultured tumor cells with interferon gamma, tamoxifen (TAM) of the TAM and with estradiol in the medium from T-lymphocytes. Additionally, higher cell viability in control samples MTS compared to control monolayer culture. In MTS significantly reduced the percentage of cells in apoptosis, regardless of the type of impact on the tumor cells. The sensitivity of cells to anticancer therapy MTS much lower as compared with monolayer cell culture. Thus, therapeutic doses of tamoxifen only cause increased proliferation and growth of MTS.
