Aim of the work: to establish the role of thyroidspecific proteins and receptor' tyrosine kinases in development of benign and malignant thyroid tumors and to determinate interralashion between expression mentioned components in thyroid cells and morphological characteristics of tumors. Methods: morphological, immunohistochemical, molecular biological, statistical. Theoretical and practical results: thyroglobuline expression was detected in all studied PTC and FA, that was correlated with histological structure of tumors. In FA, independently of there structure, increasing of NIS expression was pointed. In PTC NIS expression was decreased. Induction of RET/PTC oncogenes more frequently took place in PTC with respect to FA, and the most common oncogenes both, RET/PTC1 and RET/PTC3 were detected only in PTC. The correlation between RET/PTC expression and histological structure of FA was not established, while in PTC RET/PTC were detected more frequently in solid carcinomas. Has been proposed to modify the protocol of immunohistochemical reaction with anti-NIS antibodies, that allows to identify this protein even if its amount in cells is very low. This method give the possibility to detect NIS expression in the great majority of PTC and to point the most aggressive cancer.The results obtained will be a basis for discovering the mechanisms of malignant transformation of thyroid cells. Efficacy of introducing: the proposed method could be use for detection of aggressive phenotype of thyroid carcinomas.
