Aim of the investigations: to define the peculiarities of functioning of individual clones of murine transposon-induced pluripotent stem cells in vitro and in vivo. Theoretical and practical results: reprogramming of murine embryonic fibroblasts with Sleeping Beauty transposon-based system is achieved, the complex assessment of pluripotency characteristics, namely the change of morphology and proliferation rate, expression of pluripotency markers on mRNA and protein level, demethylation of promoters of key pluripotency genes, in vitro and in vivo differentiation potential of individual clones was demonstrated. The results on morphological and functional properties of the obtained clones can be used in further experiments on reprogramming of somatic cells of various origin and discovery of line-specific properties of induced pluripotent stem cells.
