New information concerning the effects of diphtheria toxin (DT) on the regulation of immune response has been received. Recombinant fluorescent derivatives of diphtheria toxin, recombinant transmembrane form fluorescent derivatives of proHB - EGF and soluble form of human sHB-EGF have been obtained. Test system based on the Sandwich ELISA method with the use of recombinant sHB-EGF, which allows one to detect DT at the concentration of 1.9 ng/ml has been developed. A protocol that allows receiving in preparative amounts an analog of recombinant soluble form of human HB-EGF without additional amino acid sequences (tag) has been developed. The possibility of using pEGFP-N1-proHB-EGF to study receptor function of proHB-EGF has been experimentally proved. New information about localization and molecular mechanisms of signaling of ?9(?10) nicotinic acetylcholine receptors (nAChRs) in the cells of the immune system has been obtained. Antibodies against a peptide fragment of ?9 nAChR subunit have been generated. It was shown that mouse B cells express ?9 nAChRs, the amount of which is genetically determined and increases over the B cells maturation. ?9 nAChRs are predominantly expressed in B cells of splenic marginal zone and B-1 lymphocytes of peritoneal cavity, that is a reason to consider them an evolutionary ancient immune regulatory system. ?9 nAChRs of B cells are adjacent to co-stimulatory molecule CD40, but are not connected to it physically. They negatively regulate the B cells proliferation stimulated through CD40 and are able to partially compensate the absence of ?7 nAChRs. The possibility of using mesenchymal stem cells of human adipose tissue for paracrine regulation of dermal fibroblasts of patients with diabetes mellitus type 2 has been investigated. It was shown that mesenchymal stem cells of adipose tissue of healthy people did not differ from those obtained from patients with diabetes mellitus type 2 by their adhesive and proliferative properties; increasing concentrations of glucose in the incubation medium did not affect the expression of CD44 and ICAM-1 and secretion of FGF-2 and TGF?1. New methods for estimation of ligand- receptor binding have been developed and offered to the use. An equation for determining the affinity of the antibodies, which allows to detect the presence of low affinity antibodies in the samples has been proposed. A theory has been proposed, which allows one to determine the value of the affinity constant of the antibody-antigen interaction in case, when antibodies and corresponding antigens are mixed in biological fluid and their separation into individual components is not possible because of the high lability of macromolecular complexes, the antigen concentration being unknown.
