Studying the effect of N-acylethanolamines on the viability index of normal and transformed cells in vitro under irradiation was shown that N-acylethanolamines do not influence on the proliferative activity of the irradiated cells. It was not found any significant changes on the level of apoptosis of the irradiated 1 Gy dose cells under the NAEs effect, whereas under the irradiation at a dose of 10 Gy N-stearoylethanolamine (NSE) alone showed apoptosis-stimulating action. It was shown that NAEs may modulate the activity of mitochondrial enzymes (succinate dehydrogenase and a-glycerophosphate dehydrogenase), and the phospholipid composition of both normal and transformed cells. It has been shown that the N-acylethanolamines altered the functional properties of ion channels in cardiac cells: modulated electrical excitability of the plasma membrane of neonatal cardiomyocytes, blocked sodium channels, reduced the amplitude and the initial rate of action potential rise. The mechanism of NAEs action on cardiac L-type voltage-gated calcium channels most likely involves influence on channel gating that reduces "window current" as well as partial blockade of the ion-conducting pathway that decreases current amplitude. NSE showed protective effect on the testicular tissue of rats with experimental estrogenization: normalized the testosterone level, restored pro-/antioxidative balance by modulation of antioxidant enzymes activity, prevented changes in phospholipid composition, which play an important role in the acrosomal reaction of zoosperms. NSE prevented the increase in free saturated FA pool, and decrease in the level of free unsaturated FA, mainly monoenic and dienoic FA; restored the content of docosahexaenoic acid in the rat testes that may prevent the testes dysfunction and male infertility. It has been found the protective effect of NSE on the rat testes under streptozotocin-induced diabetes. NSE application contributed the restoration of the pro-/antioxidative balance and reduction of the lipid peroxidation in the rat testes and blood plasma by restoring the activity of antioxidant enzymes, caused normalization of total and individual phospholipid content, cholesterol esters level, saturated and unsaturated FA composition in the testes of rats with diabetes. It was revealed the growth of free docosahexaenoic acid content under NSE influence. The calculated activity of the ? 9 -desaturase was restored under the action of NSE. In order to consideration the important role impaired FA composition of testes in testicular dysfunction development, we can assume that the corrective influence of NSE on the testes lipid composition may restore their functional activity. The investigation of the saturated NAEs effect on NO -synthase signaling system in rat testes under various endocrinopathies showed that NSE exerted its normalizing effect by different mechanisms. In rat testes with experimental estrogenization the growth of nitrite content under the NSE action was associated with the restoration of testosterone level. Under the type I diabetes, which was accompanied by generalized oxidative and nitrosative stress, NSE triggered a decrease of nitrite level in the rat testes. NSE increased the bioavailability of NO by its release from the deposited forms in rat testes with obesity-induced insulin resistance. The results of these studies suggest that NAEs exhibit a significant protective effect under the influence of various damaging factors, this justifies the possible use of these compounds for the correction of different pathological conditions.
