The object of investigation: oxido reductases of analytical importance: (glucose oxidase (GO) of fungi Penicillium, methylamine oxidase (AMO) isolated from recombinant yeast Saccharomyces cerevisiae, horseradish peroxidase) immobilized on nanoparticles of gold (Au-NPs) and silver (Ag-NPs). The aim of the work: a development of a new nano-sized bio-recognition elements for mediator-less biosensors on glucose and methylamin. These biosensors of “third generation” on glucose and methylamine will be constructed on the base of GO and AMO, co-immobilized with peroxidase on the surface of functionalized NPs. Synthesized bio-recognizing membranes of nano-sized range will be studied in the light of their structure and catalytic properties. The aim of the 2-nd stage investigations: studies on nanoparticles conjugation with enzymes and characterization of the obtained bionanomaterials. Investigation of GO properties which immobilized on nanoparticles. The methods of the investigation: microbiological, biochemical, physico-chemical, enzymatic. As result of the 2-nd stage: Ag-NPs and Au-NPs were synthesized and modified. Different enzymes (AMO, PO and GO) were immibilized on the surface of noble metals. The structural characteristics of the bionanomaterials were investigated. Nanosizes of synthesized bionanoparticles were proved by scanning electrone microscopy. The catalytic parameters of the immobillized enzymes GO, PO and AMO were stadied. Some kinetic properties of the solubile and immobilized forms of GO were investigated. All investigations have been carried out according to the plan. As result of the 2-nd stage study, 3 papers were accepted (in press) and 2 abstracts were published.
