It is known that disturbance of mitophagy results in the formation of active forms of oxygen in the cell, which damages intracellular material and causes the early ageing of organism (Mizushima et al., 2008). Though most of the autophagic processes are studies at the model of yeast P. pastoris, the mechanisms of mitophagy for this yeast is still unknown. The task of this stage of the project was the development of the method of the investigation of mitophagy in methylotrophic yeast P. pastoris. Omp85 is a protein, which is necessary for the biogenesis of outer mitochondric membrane of the all eukaryotic organisms. The first stage of the work was the construction of the recombinant plasmids, which carry the hybrid gene omp85 P. pastoris fused with GFP gene (coded the green fluorescent protein). The molecular analysis of the vectors was made and the new recombinant strains of yeast P. pastoris, which carry this fusion were constructed.
