An objective: The development of prototypes of amperometric and conductometric sensor analyzer systems for the quantitative determination of formaldehyde, L- and D- lactate. The purpose for period 2013: Construction of biosensor's methods for analysis of the content of L- and D- lactate - as promising biomarkers for assessing severity and the effectiveness of treatment of diabetes. Methods: microbiological, biochemical, physical, chemical, enzymatic, electrochemical, software, biosensor. An optimization of synthesis of the enzymes L-lactate: cytochrome c oxidoreductase (flavocytochrome b2, FC b2) and D-lactate: cytochrome c oxidoreductase (DLDH) in recombinant strains of yeast Hansenula polymorpha - overproducers of corresponding enzymes has been performed. The schemes of purification of FC b2 and DLDH from the H. polymorpha cells were developed and preparative quantities of highly purified enzymes have been obtained. On the base of obtained preparations of FC b2 and cell debris of DLDH overproducer, the laboratory prototypes of electrochemical biosensors for the determination of L- and D-lactate in samples of biological fluids have been developed.
