The aim of the work - to study of regional infectious agents features causing sexually transmitted diseases - Treponema pallidum and Chlamydia trachomatis for their diagnostic and therapy optimization. Methods: clinical, immunological, serological, molecular. The highly sensitive and specific molecular-genetic method of DNA of Treponema pallidum detection in clinical specimens and the verification method of DNA of Treponema pallidum detection with elimination of false-positive results were developed. The studies of typing strains of Treponema pallidum circulating in Kharkiv region were carried out on the basis of the analysis of arp and tpr gene variability. There were found on the basis of the results typing arp and tpr genes of Treponema pallidum that the subtype 6e is a dominant subtype of Treponema pallidum in the Kharkiv region. Types with 2, 6, 7, 8, 13 of 60-nucleotide repeats were found in arp gene. For tpr gene the types a, b, e, g, h, i were identified. Among the isolates examined for macrolide resistance, none had the mutation in the 23S rRNA gene. This allow to recommend usage of macrolides for the treatment of syphilis with symptoms of hypersensitivity to penicillin drugs. It is shown that in the Kharkiv region the major serotypes of Chlamydia trachomatis which were determined on the basis of the characteristics of the primary structure of MOMP distributed as follows: genotype E - (25,9 ± 3,3) % of cases, G - (25,3 ± 3,3) %, F - (21,8 ± 3,1) %, C, D and I - (5,2 ± 1,7) %, B - (4,6 ± 1,6) %, Ba, J and H - (2,3 ± 1,1) % . The mutation 972T/C in the fourth variable domain gene omp1 was found. The quick method of determination of the number of domains with tyrosine in the tarP gene is developed. It allows to have full characterization of strains Chlamydia trachomatis circulating in the Kharkiv region.It was found that the Chlamydia trachomatis with 2 or 3 tyrosine domains in the tarP gene are revealed in the Kharkiv region more often. The variants with 6 or more tyrosine domains are revealed quite often as well.
