Methods of investigation - novel methods of the computer simulation of a high level of complexity, in particular quantum-mechanical (QM), and Bader's topological analysis of the electron density (QTAIM). By combining QM and QTAIM methods of investigation and by using the methodology of the scans of the basic physico-chemical characteristics of the double proton transfer along the internal reaction coordinate, for the first time we have elucidated physico-chemical mechanism of the mutagenic tautomerisation of the Gua·Thy and Ade·Cyt base pairs. For the first time a new physico-chemical mechanism of the origin of the spontaneous transitions has been proposed and substantiated. It consists in the tautomerisation of the pyrimidine bases of the wobble Gua·Thy and Ade·Cyt base pairs with their transformation into the Gua*·Thy and Ade·Cyt* mispairs, respectively, induced by the interaction with the recognition center of the correct pairs of the nucleotide bases of the replicative DNA-polymerases.
