The study shows new ITSNs protein-protein interactions which were confirmed in vitro and in vivo. Specific phosphorylation of ITSN2 mediates its recognition by SH2 domain-containing proteins and could be linked with function divergence of intersectins. A role of ITSN2 in regulation of actin remodeling during embryogenesis was shown. Analysis of expression of short and long ITSN2 isoforms in 71 sample of breast cancer indicates correlation between low expression level of ITSN2 short isoform with HER2/neu-positive status of tumour and with metastases in the lymph nodes in patients with ER/PR-positive tumours patients, which can be one of prognostic markers of the disease.
