It was shown that MPTP opening leads to the increase in transmembrane proton exchange due to the increase in K+ and Ca2+-cycling in mitochondria. The increase in potential-dependent potassium uptake results in the activation of K+/H+-exchange and the increase of proton entrance into matrix. Reciprocal dependence was observed between the increase in K+/H+-exchange and the decrease in MPTP activity due to the mitochondrial K+ATP-channel activation. The activation of transmembrane ion exchange under simultaneous opening of MPTP and K+ATP-channel leads to the uncoupling of respiratory chain and potential-independent decrease of ROS production. It was summarized that the mechanism of MPTP inhibition under K+ATP-channel opening comprises the activation of K+/H+-exchange and the suppression of ROS production.
