Research aim was to study the effect of initial parameters of the medium, amphiphilic compounds and cryoprotectants on sensitivity of erythrocytes of different types of organisms to changes in osmotic parameters of medium, cooling and freezing. In the work there were used contemporary research methods: spectrophotometry, pH-metry, osmometry, ionometry, microhematocrit method, chemical modification of cells, freezing in liquid nitrogen, hypothermic storage of cells, morphometry. Results and their novelty. Research results can be used to decode the mechanism of effect of cryopreservation factors on mammalian erythrocytes. The obtained results testify to the fact that protective effect of representatives of different classes of amphiphilic compounds under conditions of osmotic and temperature stresses is stipulated with the ability of alkyl sulphates to perturbate a membrane during building-in and to prevent the rise in transmembrane microdefects up to the size of hemolytical pore. There was established the fact that inclusion into the medium with polymer cryoprotectant of penetrating one led to weakening of erythrocyte dehydration and effect of hypertonic stress on membranes when freezing. The results of these studies allow the conclusions that morphological response might be one of the types of general reaction of cells to changing ionic environment, and the most optimal for hypothermic storage of erythrocytes are non-electrolyte incubation media with neutral values of pH.
