The purpose of the study is to develop high effective methods for the synthesis of molecular various nitrogen containing heterocyclic systems and to detect among them with using in silico and in vitro molecular screening of high research capacity compounds that can inhibit the development of metabolic abnormalities linked with type 2diabetes and to determine their specific effects on experimental models. Methods of the study: organic synthesis, physical and chemical analysis, biochemical, pharmacological and molecular-biological. A series of new compounds of pyrazolo [3,4-b] quinoline-5-ones and thiazine classes with established structure were synthesized. Pharmacological screening in vitro and in vivo of a number of heterocyclic compounds tetrahidropirazolo [3,4-b] quinoline-5-ones and thiazine derivatives was carried out. The results of primary screenings of 17 different structural types compounds on the presence of hypoglycaemic and pro/anti-oxidative properties were found promising substances 2418, 2419 as potentive antidiabetic compound. The investigations of the influence of heterocyclic compounds 2418 and 2419 for type 2 diabetes mellitus, caused by the introduction of streptozotocin on the background of high fat diet in rats, showed their ability to reduce disglycaemic parameters, insulin resistance and oxidative stress. The results of the study of apoptosis manifestations were demonstrated the presence of positive impact of oral use of 2418 and 2419 compounds for a total survival rate of liver cells of diabetic animals.
