It was shown that the use of pSUP5011::Tn5 vector is more effective than the pSUP2021::Tn5 vector for insertion of Tn5-transposon in the genome of B.japonicum slow growing nodule bacteria and allows to obtain a wide range of Tn5-mutants. It was selected the high effective Tn5-mutantas of soybean, alfalfa and clover rhizobia wich on the main parameters of efficiency of interactions with plant and on resistance to drought exceed the parent strains by 10-30%. It was shown that obtained rhizobia are able to keep their symbiotic properties, to introduce successfully in the soil and form effective symbiotic systems with host plants, ensuring high and stabile yields. It was established thet the seed inoculation leads to the expression of genes which are responsible for the symbiosis formation and the level of the expression is independent of the properties of symbiosis microorganisms. It promotes the increase of total protein content in plant roots infected by active rhizobia. It was proved that the modeling of rhizobia physiological activity by the adding of the biological active component of protein nature - lectin- ensures their high effectivity compared to standard strain. Modified by homological lectin rhizobia for manufacturability comply with requirements that apply to microorganisms used in the biopreparate production. Based on the results of researches it was developed the bacterial preparate Rhyzostym which use is a highly effective means to improve of nitrogen nutrition of legume throught the use of atmosphere molecular nitrogen, increase in plant resistance to the action of environmental stress, which in turn quarantees a high and stable yields, in particular, under unfavorable environmental conditions.
