Karazin popup.description2 3- and 24-month-old Wistar male rats and isolated hepatocytes as well as skeletal muscle, brain cortex, hippocampus, hipotalamus, reticular formation of un-treated or calorie-restricted animals or emotional stress-treated rats were used. Spectrophotometric, radioisotope, electro-physiological, chromatographic methods and "open field" test and shuttle box were used in the work. The aim of the work was to study the role of different pathways of sphingolipid turnover in the regulation of thyroxin and insulin signal transduction and the calorie restriction effect on sphigolipid and cardiolipin metabolism in the brain, heart and liver. Moreover, effect of inhibition of neutral sphingomyelinase activity with N-acetylcysteine on the behavioral and cognitive functions of old rats and bioelectrical activity of brain structures of rats with high and middle levels of movement activities under emotional stress have been studied. It has been determined by means of specific inhibitors of sphingolipid synthesis and degradation that activation of ceramide synthesis de novo and sphingomyelin degradation in the studied tissues was associated with the insulin and thyroxin resistance development in the target cells at old age. Emotional stress increased ceramide content in the hippocampus, which followed with significant changes of rat behavior in the "open field" and electric activities of brain structures.
