During the reporting period, we studied the effect of mesenchymal stem cells (MSCs) on the development of the tumor cell population mammary adenocarcinoma at monolayer steroid and growth. According to the literature mesenchymal stem cells are able to inhibit the growth of tumor population and "normalize" the phenotype of tumor cells. Research Facility: mesenchymal stem cells from bone marrow and human tumor cell line MCF-7. In this work were used immunocytochemistry, flow cytometric and molecular biological techniques, biochemical and statistical analysis methods. The aim of the study was to characterise the effect of human bone marrow MSCs proliferation, survival, and the receptor profile of the ability to form multicellular spheroids tumor cells in vitro. The objectives of the study was to determine the kinetic parameters of the growth of human tumor cells in culture for co-cultured with MSCs human bone marrow assessment of the level of expression of some oncogenic markers of tumor cells by the bone marrow MSCs man and a comparative analysis of the above parameters at different stages of cultivation in monolayer and spheroid culture. Comparison was made on the level of expression of estrogen receptors, epidermal growth factor receptor, cytokeratins and E-cadherin. The Results. According to the study of human bone marrow MSCs have a cytostatic effect on the tumor population. It turned out that the non-contact co-cultivation of human bone marrow MSCs with MCF-7 percent of EGF-R+ cells increased by 18%, the number of AE1/AE3+ cells increased by 54% and save the number of Pan+ cells at the level of 100%. According to the authors of the percentage of cells expressing E-cadherin after coculture with human bone marrow MSCs was reduced by 32%, ER + cells and increased by only 8%. According to the results of experiments, it was found that the introduction of cytosolic CK inhibited tumor growth kinetics by 37%. In 10% of the animals developed tumors at all. At the same time there is a relative normalization of blood biochemical parameters in experimental animals (total protein, albumin, ALT, AST, LDGG). The findings have both scientific and practical significance. From a scientific point of view was to study the fundamental mechanisms of interaction of the tumor cell population and the microenvironment. When non-contact with the cultured here are messengers humoral signals (interleukins, cytokines, growth factors), and mediators - tumor cell receptors, which in turn trigger a cascade of proliferative, adhesion and migration processes. From a practical point of view, determine the conditions of antitumor action of human bone marrow MSCs, especially at the level of an animal model will allow to develop effective anti-tumor agents based on cytosolic human stem cells without the risk of inducing tumor progression and cell contamination.
