Research report: p. 53, fig. - 5, Table. - 2, Photo - 3, reference sources – 87. Research Facility: tumor spheroids of MCF-7 cells, interferons alpha, gamma, estradiol, tamoxifen. In this work were used immunocytochemistry, flow cytometric and molecular biological techniques, biochemical and statistical analysis methods. The aim of the study was to characterize the influence of humoral factors in the microenvironment of the proliferation, survival, metastatic potential of tumor cells in multicellular spheroids. Determination of proliferative and morphological characteristics of spheroids and monolayer culture in different conditions microenvironment. The objectives of the study was to generate multicellular tumor spheroids (МTS) breast cancer cells. Determine proliferative and morphological characteristics under various conditions МTS microenvironment. As a result, performance has been demonstrated that a medium from T-lymphocyte line MT-4 significantly stimulated growth of live tumor cells in suspension or in adhesion. But in the presence of fibroblast-growth environment of the tumor cell population is inhibited as a suspension or in adhesion. Moreover a large number of tumor spheroids formed, adding to the medium of the T-lymphocytes. And the least when co-cultured with fibroblasts. The obtained results may indicate a more EGF-like secretory capacity monolayer culture of MCF-7 cells (15.7 ng), compared with a spheroid (6.8 ng). Scientific and practical significance. Thus, in the result of the work was confirmed by the assumption of the antitumor effect of stromal microenvironment. And the threat of stimulating tumor growth secretory signals from T-lymphocytes. It is particularly important confirmation of the data on the culture of tumor microaggregates model of micrometastases.
