Purpose: creation of the device for quick freezing, to study the effectiveness of using this device for the survival of MCF-7 cells in a medium with different cryoprotectants Methods: cryoprotective solutions (CR) consisted of the culture medium, serum, DMSO, sucrose, glycerol, ethylene glycol, polyethylene glycol different ratios. When you add the CR using five different protocols at low and ultra-fast freezing using the prototype of the device. The results and their novelty: in accordance with the results obtained, the protocol 2 was more effective than one protocol, for all compositions of cryoprotectants. This can be explained by a gradual decrease in temperature of the cell suspension with delayed addition of a penetrating cryoprotectant DMSO. It is known that at lower temperatures the cell to +4 ° C in cell metabolic processes are suspended. In this state, the cell organelles and enzyme systems are less exposed to the harmful effects of DMSO, which increases the total survival cells. Of the five compositions studied cryoprotective solutions greatest protective effect on the cells had a solution which includes in its membership 20% sucrose and 5% DMSO. The cooling rate at 0- -40 ° C temperature range was 159.6 deg/min. From -40 ° C to -180 °C, the rate of 560 deg/min. In the future, the results will be used to determine an effective method of ultrafast cooling and storing cells.
