Aim of the study: use of technologies of gene- therapeutic correction of insulin deficiency by administration into the body of a molecular construction with a functionally active gene of preproinsulin. Methods and equipment: biochemical, immunoenzymic methods; Super GL analyzer, NanoDrop “Jen Way” spectrophotometers, Accu Chek “Active” glucometer. Results: a study of the effects of administration of DNA recombinant molecules containing a target gene of human preproinsulin on streptozotocin diabetes models to C57BL/6j mice and Wistar rats, has shown a significant decrease in hyperglycemia in diabetic animals. Theoretical and practical novelty: the data obtained represent a new and important information that will be taken into account in further investigations aimed at determining conditions of an effective expression of insulin gene in hepatic cells of experimental animals with induced insulin-dependent diabetes mellitus. Novelty and advantages over analogues: For the first time, the effects have been studied, of the administration of DNA recombinant molecules containing a target gene of human preproinsulin, in mice and rats with different duration of streptozotocin diabetes. Based on an analysis of the administration into liver parenchyma of diabetic animals of different concentrations of a molecular construction with a functionally active gene of preproinsulin, optimum doses of DNA preparations have been determined, that lead to a significant and durable (up to two months) decrease in hyperglycemia. Efficacy of introduction: introduction of the results of study will include publication of articles in Ukrainian and foreign journals, by presentations at scientific congresses and conferences.
