Aim - Methods - radiobiological, biochemical, microscopic, spectrophotometric, chromatographic and statistical techniques as well as a method atomic absorption spectrometry. Dynamics of changes of biochemical activity and composition of semen in rabbits after their total irradiation in doses 1.0-7.0 Gy was revealed. X-ray exposure in doses 2.0 Gy, 5.0 Gy and 7.0 Gy has dose-dependent suppressive effect on prostate, seminal vesicles and epididymis activity which was manifested by decrease of fructose, citrate, L-carnitine, а-tocopherol, ascorbate, free thiols and prostaglandin levels in rabbit's semen. Later, 90 days after exposure by 2.0 Gy and 5.0 Gy partial normalization of some parameters was marked, and its length depended of exposure dose. Experiments performed in model system using experimental animals - rabbits, made possible selection of the most sensitive to ionizing irradiation parameters of seminal fluid. In comparative randomized cohort studies down-regulation of ascorbate and glutathione, free carnitine and thiols, citrate and zinc in sperm of male donors from polluted by radiation territories, e.g. Zhytomirska oblast, was shown. Appearance of acrosomal enzymes - acrosine, hyaluronidase and acid phosphatase in seminal fluid of men living in polluted by radiation territories of Ukraine was shown for the first time and it suggest acrosome damage in their spermatozoa. However, elevation of а-tocopherol, fructose, prostaglandin F2а levels, enhancement of lipid peroxidation and decreased prostaglandin E concentrations were observed in their seminal fluid also.
