Objective: To develop the technology to produce antigen for complement fixation reaction for the diagnosis of glanders horses, and determine the specificity and sensitivity of antigen produced in setting complement fixation reaction with serum of horses, selected from different regions of Ukraine. Methods and apparatus: Cultivation of glanders pathogen Burkholderia mallei Bogor, Mukteswar, Zagreb, making hoe antigen for complement fixation reaction, determine the sensitivity and specificity obtained antigen. The result: A hoe laboratory sample antigen for complement fixation reaction with strains of B. mallei Bogor, Mukteswar, Zagreb, studied the performance of the diagnostic sensitivity and specificity. Key Features and performance: Antigen hoe for CFR received by heat inactivation agar culture strains of glanders B. mallei Bogor, Mukteswar, Zagreb and stabilize the resulting suspension 0.5% solution of phenol. The antigen was studied indicators of diagnostic sensitivity and specificity. Diagnostic sensitivity was defined as the proportion of positive reference sera that react positively in CFR. Diagnostic Specificity was defined as the proportion of negative reference sera, which in CFR react negatively. Found that sensitivity and specificity obtained for CFR antigen hoe not inferior, used for serological diagnosis of glanders in Ukraine. Recommendations for the use of results: Based on the results preparing proposals for amendments to the guidelines for the diagnosis of glanders and instructions on prevention and control of glanders animals. Proposals are to implement long-term complement fixation reaction (LT-CFR) in order to increase the sensitivity of the method applied micromethod setting CFT, lighting and production method validation hoe antigen for CFR. Range of application: veterinary medicine, laboratory diagnostics. Conclusion: The method for manufacturing an antigen CFR hoe and prepared proposals to improve the method of setting CFR for serological diagnosis of glanders animals.
