But detection of scFv and their complexes with antigens is problematical: prevailing systems for detection have some drawbacks and are not universal We constructed fusion proteins, which contain sequences of scFv and staphylococcal protein A fragment or fluorescent protein mCherry. It was shown, that scFv fused with protein A and their complexes with antigens were effective recognized by labelled immunoglobulins and that yield of active functional fusion proteins in a similar expression system was significantly higher than the yield of free scFv. ScFv to growth factor HB-EGF fused to mCherry were functionally active in flow cytometry and immunofluorescent microscopy.
