Four microsatellite loci with valid PCR products were identified. Polymorphisms were estimated in populations of four species of the genus Vincetoxicum from the south east Ukraine. Intraspecific variation by microsatellite loci is close to interspecific one, and different predominant alleles are present in some populations. We revealed the founder or bottleneck effects in populations. Some loci and genotypes, useful for Vincetoxicum species differentiation were identified (V. tauricum Pobed. - Vinc123134/134, Vinc124323/410; V. intermedium Taliev - Vinc123142/142, Vinc124317/317). We analyzed ISSR polymorphisms by seven primers in Astragalus L. distributed in Ukraine. The study has shown applicability of ISSR loci to species differentiation in this genus. Genetic diversity of study populations of four Astragalus species from the south east Ukraine was described. Intraspecific variation of study populations is close to interspecific one that is indicative of the gene flow or population bottlenecks. Based on molecular genetic markers, a general framework of biodiversity analysis useful for rare and endangered species' taxonomy studies, restoration and conservation is introduced.
