Dokuchaev Kharkiv National Agrarian University popup.description2 The involvement of nitric oxide as a signal mediator and its interaction with reactive oxygen species (ROS) and calcium ions in the process of induction of heat-resistance of wheat (Triticum aestivum L.) seedlings by heat hardening, exogenous nitric oxide donor sodium nitroprusside (NNP), calcium salt (CaCl2), hydrogen peroxide, and 24-epibrassinolide phytohormone has been studied. The effect of transient increase in nitrogen oxide content in roots of seedlings exposured to hyperthermia and treated by all these compounds was founded. This effect eliminated by the action of antioxidants ionol (BHT) and dimethylthiourea, calcium channel blockers EGTA and lanthanum chloride, as well as inhibitors of phospholipase C neomycin, and thus NO maintaining was dependent on the formation of ROS and calcium status of cells. These compounds significantly inhibited the development of heat-resistance of wheat seedlings caused by exogenous nitric oxide. The involvement of nitric oxide in the effects of hydrogen peroxide in the development of heat-resistance of seedlings was found too. Leveling by PTIO scavenger of nitric oxide and NO-synthase inhibitor L-NAME of positive effects of brassinosteroid in conditions of hyperthermia suggests involvement of nitric oxide in the regulation of gene expression by 24-epibrassinolide. In order to establish the theoretical foundations of NNP use in crop we studied the effect of spraying with NNP solutions on the physiological state and productivity of millet (Panicum miliaceum L.) plants in field experiments under adverse conditions. NO donor treatment of plants in optimal concentrations was to mitigate the negative impact of adverse soil conditions, high temperature and lack of water, as evidenced by preservation of high content of photosynthetic and flavonoid pigments in flag leaf and higher plant treated with NNP productivity in comparison with control. Thus, in this work the induction by nitric oxide of heat resistance of plants in the early stages of development was investigated, a close functional interaction of NO with other signaling mediators, in particular with ROS and calcium ions was proven, the participation of NO in implementing the physiological effects of heat hardening and exogenous brassinosteroid was founded, and possibility of using NO donor in adverse field conditions for improving the physiological state of plants has been shown. Results of the research are used in teaching the "Plant Physiology" course in V.V. Dokuchaev KhNAU. They can also be used by research institutions in developing new approaches to induce plant resistance to abiotic stressors.
